Target Information

PTEN was identified as a tumor suppressor frequently mutated in various cancers. The PTEN gene encodes a phosphatidylinositol-3,4,5-trisphosphate 3-phosphatase containing a tensin-like domain and a catalytic domain similar to dual-specificity protein tyrosine phosphatases. Unlike most protein tyrosine phosphatases, PTEN preferentially dephosphorylates phosphoinositide substrates. It negatively regulates intracellular levels of phosphatidylinositol-3,4,5-trisphosphate and functions as a tumor suppressor by inhibiting the AKT/PKB signaling pathway. PTEN modulates the AKT-mTOR signaling pathway, influencing neuron integration, dendritic development, and synapse formation during adult neurogenesis. Additionally, PTEN may negatively regulate insulin signaling and glucose metabolism in adipose tissue. The nuclear monoubiquitinated form shows higher apoptotic potential, while the cytoplasmic nonubiquitinated form exhibits reduced tumor suppressive ability.
