Product Specific Information

This product consists of two fluorochrome-conjugated monoclonal antibodies to identify CD4+CD25+ human peripheral blood cells.

• RPA-T4 monoclonal antibody (mouse IgG1, kappa) reacts with human CD4, a 59 kDa cell surface receptor expressed by most thymocytes, a subset of mature T helper cells, and at low levels on monocytes.
• BC96 monoclonal antibody (mouse IgG1, kappa) reacts with human CD25, the 55 kDa interleukin-2 receptor alpha chain (IL-2Rα).

This cocktail can be used for flow cytometric detection of human regulatory T cells, which also express Foxp3.

Applications Reported: Flow cytometric analysis.
Applications Tested: Pre-titrated and tested for flow cytometric analysis of normal human peripheral blood cells.
Use 20 µL per test (contains 1 µg Anti-Human CD4 (RPA-T4) FITC and 0.125 µg Anti-Human CD25 (BC96) APC).
A test is defined as the amount (µg) of antibody that will stain a cell sample in a final volume of 100 µL.
Cell number should be determined empirically (10⁵–10⁸ cells/test).

Target Information

CD4:
Involved in recognition of type II MHC antigen and serves as a receptor for HIV. Expressed on most T helper cells and thymocytes. The cytoplasmic portion associates with p56lck tyrosine kinase. CD4 expression is common in human lymph nodes and tonsils.

CD44:
A type I transmembrane glycoprotein (Phagocytic Glycoprotein-1, HCAM), receptor for hyaluronate, existing in multiple isoforms due to RNA splicing. Involved in leukocyte adhesion to endothelial and stromal cells.

CD25 (IL2RA):
Forms part of the high-affinity IL-2 receptor complex with IL2RB and IL2RG. Homodimeric alpha chains yield low-affinity receptors; beta chains yield medium-affinity receptors. Soluble IL2RA arises from extracellular proteolysis. Mutations in IL2RA are linked to interleukin-2 receptor alpha deficiency.
