Target Information

Human Rad18 (hRad18) is a homologue of the S. pombe Rad18. In yeast, Rad6 and Rad18 play a crucial role in post-replication repair, facilitating gap-filling of the daughter strand during replication of damaged DNA. The Rad6–Rad18 interaction is critical at an early step during lesion bypass. In vivo, Rad18 forms a tight complex with Rad6 through a conserved ring-finger motif. The Rad18–Rad6 complex binds to damaged sites via the single-stranded DNA binding property of Rad18, followed by degradation of stalled replication components through the ubiquitin-conjugating activity of Rad6.
Human Rad18 maps to chromosome 3p24–25, a region often deleted in lung, breast, ovary, and testis cancers. The human RAD18 gene encodes a 495 amino acid protein with a molecular weight of approximately 56 kDa and is ubiquitously expressed in various human tissues.
