Target Information
Hyaluronic acid (HA) is a high molecular weight, unbranched polysaccharide found in organisms from bacteria to mammals and is a key component of the extracellular matrix. It is composed of alternating glucuronic acid and N-acetylglucosamine residues linked by β-1,3 and β-1,4 glycosidic bonds. HA is synthesized by membrane-bound synthases at the inner surface of the plasma membrane and extruded into the extracellular space.
HA functions in space filling, joint lubrication, and as a scaffold for cell migration during wound healing and tissue repair. Changes in serum HA concentration are associated with inflammatory and degenerative joint diseases such as rheumatoid arthritis. The interaction between HA and the leukocyte receptor CD44 plays a crucial role in tissue-specific leukocyte homing, and overexpression of HA receptors has been linked to tumor metastasis.
HAS1 (Hyaluronan Synthase 1) is a member of the vertebrate gene family encoding hyaluronan synthases and shares significant homology with the hasA gene product of Streptococcus pyogenes.
