• Immunohistochemistry (PFA fixed): Incubate antibody for 30 minutes at room temperature.
• For formalin-fixed tissues: Heat tissue sections in 10 mM Tris with 1 mM EDTA (pH 9.0) for 45 minutes at 95°C, then cool at room temperature for 20 minutes.

Target Information

ICAM-1 (CD54) is an 85–110 kDa single-chain type I integral membrane glycoprotein composed of five immunoglobulin superfamily repeats in its extracellular domain, a transmembrane region, and a cytoplasmic domain.
It has seven potential N-linked glycosylation sites and shares significant sequence homology with ICAM-2 (CD102) and ICAM-3 (CD50).
ICAM-1 binds to integrins such as CD11a/CD18 (LFA-1) and CD11b/CD18 (Mac-1), and serves as a receptor for Rhinovirus.
It is expressed on activated endothelial cells, epithelial cells, fibroblasts, chondrocytes, and various immune cells including B and T lymphocytes, monocytes, macrophages, dendritic cells, and neutrophils.
Expression levels increase upon inflammation, and soluble ICAM-1 can be detected in plasma, often elevated in inflammatory conditions.
