Target Information
The Ras superfamily of small GTP-binding proteins are critical mediators of diverse cell signaling pathways, including those leading to proliferation, cytoskeletal organization, and secretion. The counter-conversion between the active GTP-bound and inactive GDP-bound forms is regulated by proteins that alter GTPase activity or GDP/GTP exchange rates.
Guanine nucleotide-releasing factors (GRFs) increase GDP dissociation, while GDP-dissociation inhibitors (GDIs) decrease it. The Rho GDI subfamily includes Rho GDIα, Ly-GDI (Rho GDIβ, previously GDI/D4), and Rho GDIγ. These proteins interact with several Ras-like GTP-binding proteins such as Rho A, Rho B, Rac, and Cdc42. Ly-GDI is expressed mainly in hematopoietic cells, particularly B and T lymphocyte cell lines.
