Product Specific Information

The GK1.5 monoclonal antibody reacts with the mouse CD4 molecule, a 55 kDa cell surface receptor expressed by thymocytes, mature T cell subsets, and dendritic cells. CD4 binds to MHC class II molecules on antigen-presenting cells and is essential for T cell development and function. In T cells, CD4 associates with protein tyrosine kinase p56lck via its cytoplasmic tail. Binding of GK1.5 is blocked by RM4-5.

• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported:
Suitable for flow cytometric analysis. Internal tests show NovaFluor Yellow 570 non-specifically stains B cells in SJL mice but not in BALB/c or C57BL/6 strains.

Applications Tested:
Validated for flow cytometric analysis of mouse splenocytes. Recommended use ≤ 0.2 µg per test (100 µL final volume). Cell count range: 10⁵–10⁸ cells/test. Optimize antibody titration for best performance.

Compatibility Notice:
NovaFluor dyes are incompatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI). Use Invitrogen LIVE/DEAD Fixable Dead Cell stains instead.

Buffer Recommendation:
CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining.

• Add 5 µL per sample to antibody cocktails before combining with cells.
• For single-color controls: 5 µL per 100 µL cell sample (10³–10⁸ cells).

Based on Phiton™ technology utilizing nucleic acid dye structures engineered for optimized fluorescence with reduced spillover and spread.

Target Information

CD4 antigen is a co-receptor for MHC class II molecules and HIV. Expressed mainly on T helper cells, monocytes, macrophages, dendritic cells, and certain tissues (thymus, lymph nodes, spleen, tonsils, brain, gut). CD4 participates in early T-cell activation and may mediate neuronal damage in immune-related CNS diseases. Multiple alternatively spliced transcripts are known (RefSeq, July 2017).
