Target Information
NEK4 is a serine/threonine kinase involved in signal transduction and cellular communication. It acts primarily on threonine residues and is expressed in most primary carcinomas. Protein kinases transfer phosphate groups from ATP to substrate proteins, mediating signal transduction and regulating cellular metabolism, transcription, cell cycle progression, cytoskeletal rearrangement, apoptosis, and differentiation.
The protein kinase family includes over 500 gene products, classified into 8 major groups based on sequence comparisons of tyrosine (PTK) or serine/threonine (STK) kinase catalytic domains. The STE group (homologs of yeast Sterile 7, 11, 20 kinases) contains 50 kinases related to MAPK cascade families (Ste7/MAP2K, Ste11/MAP3K, and Ste20/MAP4K). MAP kinase cascades, including MAPK and upstream kinases (MAPKKs), are well characterized in yeast pheromone response pathways, where pheromones activate MAPK signaling through Ste surface receptors.
