Target Information
The E3 ubiquitin-protein ligase NEDD4 is a Ubl conjugation pathway protein containing one HECT (E6AP-type E3 ubiquitin-protein ligase) domain and four WW domains. It participates in VEGFR-2/KDFR degradation independently of its ubiquitin-ligase activity and acts as a potential proto-oncogene by negatively regulating PTEN via ubiquitination. NEDD4 is also involved in viral budding and regulation of epithelial Na⁺ channels, influencing arterial hypertension. It localizes to the sarcolemmal region of muscle fibers, with increased expression observed in unloaded and denervated muscles, leading to decreased Notch1 expression and downregulation of Notch-1 gene expression.
