Target Information

The tumor suppressor protein p53 is a sequence-specific transcription factor activated by cellular stress. It mediates cell cycle arrest or apoptosis in response to DNA damage or pyrimidine nucleotide starvation. Upon stress, p53 activates transcription of specific genes such as p21waf1, leading to G1 or G2/M arrest. Structurally, p53 includes an N-terminal transactivation domain, a central DNA-binding domain, an oligomerization domain, and a C-terminal regulatory domain.

Phosphorylation at Ser15 is critical for p53 activation and stabilization, while phosphorylation at Ser392 is associated with tumor formation. p53 plays a key role in preventing proliferation of damaged cells and acts as an anticancer factor. It is also linked to aging, oxidative stress, and cognitive function through its regulation of long-term potentiation (LTP).

In normal cells, p53 levels are low, but in transformed cell lines, expression is elevated and contributes to malignancy. Mutations in the TP53 gene, common in many human cancers, disrupt DNA binding and abolish tumor suppressor activity. Germline TP53 mutations are also found in cancer-prone families such as those with Li-Fraumeni syndrome.
