Immunohistochemistry (PFA fixed): incubate antibody for 30 minutes at room temperature.
Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA (pH 9.0) for 45 minutes at 95°C, followed by cooling at room temperature for 20 minutes.

Target Information

Methyl-CpG-binding domain protein 1 (MBD1) is a member of a family of mammalian proteins that specifically bind methylated regions of DNA. It functions as a transcriptional repressor by binding CpG islands in promoters where cytosine is methylated at position 5 within CpG dinucleotides. Binding is abolished by the presence of 7-mG, produced by DNA damage via methylmethanesulfonate (MMS).
MBD1 plays a role in gene silencing by recruiting AFT7IP, which in turn recruits factors such as the histone methyltransferase SETDB1. Isoform 1 and isoform 2 can also repress transcription from unmethylated promoters.
