Target Information
CD28 antigen is a 44 kDa disulfide-linked homodimeric T cell-specific surface glycoprotein. It belongs to the immunoglobulin superfamily and is constitutively expressed on most peripheral blood T lymphocytes. CD28 acts as a critical costimulatory receptor, providing the second activation signal to T cells by binding CD80 and CD86 on antigen-presenting cells.
In addition to its co-stimulatory role, CD28 prevents T cells from entering an anergic or apoptotic state. In murine peripheral lymphoid organs and blood, all CD4+ and CD8+ T cells express CD28. Expression is highest on immature thymocytes and decreases in mature T cells. CD28 dysfunction is associated with diseases such as mycosis fungoides and Sezary’s disease.
