Target Information
STAT4 was originally identified using degenerate primers complementary to conserved regions of other STAT proteins. STAT4 shares similarity with STAT1 (52%) and STAT3 (47%) and can be tyrosine phosphorylated by Jak1 or Jak2. It forms homo- and heterodimers with related STAT family members and binds the IFN-gamma activated site (GAS). Serine phosphorylation is also required for maximal transcriptional activity.
STAT4 expression is mainly found in the thymus, spleen, and testis. It is activated by interleukin 12 (IL-12), which is essential for IFN-gamma production and Th1 immune response development. STAT4-deficient mice show impaired Th1 development and enhanced Th2 differentiation. In viral infection, IFN-α/β activation of STAT4 is required for IFN-γ production.
