Target Information

Retinoic acid-inducible gene I (RIG-I) is a pattern recognition receptor (PRR) involved in the detection of viral double-stranded RNA (dsRNA). Together with MDA5, RIG-I activates innate immune responses by recognizing viral dsRNA. Both proteins are RNA helicases with overlapping and distinct roles.
RIG-I is activated by dsRNAs lacking a 5'-triphosphate end and short dsRNAs, while MDA5 responds to long dsRNAs. Once activated, both signal through IPS-1 to activate NF-kappaB and IRF-3 transcription factors, leading to apoptosis, cytokine signaling, and inflammation.
RIG-I plays a critical role in immune signaling against influenza A/B, human respiratory syncytial virus, paramyxoviruses, Japanese encephalitis virus, and West Nile virus. MicroRNA-146a negatively regulates RIG-I-dependent antiviral responses by targeting TRAF6, IRAK1, and IRAK2.
Recent findings also suggest that RIG-I can detect cytosolic DNA through RNA polymerase III activity.
