Product Specific Information

Description:
The MEL-14 monoclonal antibody reacts with mouse CD62L, a 76 kDa member of the selectin family. CD62L is expressed on neutrophils, monocytes, and subsets of T, B, and NK cells. It binds glycosylated, fucosylated, sulfated sialylated glycoproteins including CD34, glycam-1, and MAdCam-1, mediating lymphocyte rolling on activated endothelium and homing to high endothelial venules (HEV) of peripheral lymphoid tissues.

Applications Reported:
Reported for use in flow cytometric analysis.

Applications Tested:
Tested by flow cytometry using mouse splenocytes. Recommended ≤0.125 µg/test (100 µL final volume). Cell number: 10⁵–10⁸ cells/test. Optimize antibody titration for best performance.

Fluorochrome Information:
APC-eFluor 780 emits at 780 nm, excited with the Red laser (633 nm). Ensure instrument compatibility.

Light Sensitivity:
This tandem dye is sensitive to photo-induced oxidation. Protect vial and stained samples from light.

Fixation:
Samples can be stored in:

• 1-step Fix/Lyse Solution (Product #00-5333)

Store up to 3 days in the dark at 4°C with minimal effect on brightness and FRET efficiency.

Target Information

Human CD62L (L-Selectin) is a 74–95 kDa glycoprotein adhesion receptor. It includes:

• Aminoterminal C-type lectin binding domain
• EGF-like domain
• Two short consensus repeat (SCR) sequences
• Transmembrane and cytoplasmic regions

CD62L is constitutively expressed on most leukocytes, including lymphocytes (except some memory T cells), monocytes, and polymorphonuclear cells.
