Target Information

CD20 is a non-glycosylated surface phosphoprotein with a molecular weight of 33–37 kDa depending on phosphorylation. It is expressed on mature and most malignant B cells, a subpopulation of T lymphocytes, and follicular dendritic cells. CD20 expression is synchronous with surface IgM and regulates calcium conductance, cell cycle progression, and B-cell proliferation. It associates with lipid rafts depending on extracellular triggering and BCR aggregation. Upon receptor ligation, BCR and CD20 colocalize and then dissociate before BCR endocytosis, while CD20 remains at the cell surface. CD20 is a key target for antibody-mediated B-cell depletion in malignancies, as it remains on the membrane after treatment. Dysfunction in CD20 is linked to Ms4a1-related common variable immune deficiency.
