Background
Type IV collagen은 basement membrane의 주요 구조적 성분으로, 3개의 α 서브유닛으로 구성된 다중 단백질입니다. 각각의 α 서브유닛은 6개의 서로 다른 유전자(alpha 1~6)에 의해 암호화되며, 서로 결합해 triple helix 구조를 형성합니다. COL4A3 유전자는 α3 서브유닛을 암호화하며, Goodpasture 증후군에서 autoantibody가 alveoli와 glomeruli의 basement membrane에 결합합니다. 이러한 항체의 주요 epitope은 단백질의 C-말단 비콜라겐 영역에 위치하며, 해당 영역은 병리적 상태에서 특정 kinase에 의해 인산화됩니다. 또한 COL4A3 유전자는 Alport 증후군의 열성 유전형과도 관련되어 있으며, 주요 변이는 동일한 C-말단 영역을 암호화하는 엑손 내에 존재합니다.
