Product Specific Information

The OKT3 monoclonal antibody reacts with an epitope on the epsilon-subunit within the human CD3 complex. It has potent immunosuppressive properties and has been effective in treating renal, heart, and liver allograft rejection. CD3 is expressed by thymocytes and all mature T cells, and its crosslinking initiates cellular activation and proliferation. Clones OKT3 and SK7 recognize different epitopes.

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated for flow cytometry using human peripheral blood cells at 5 µL (0.25 µg) per test.

Super Bright 645 is a tandem dye excited by the violet laser (405 nm) and emits at 645 nm. Recommended filter: 660/20 bandpass. Ensure instrument compatibility.

When using multiple Super Bright dye-conjugated antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize non-specific polymer interactions.

Light Sensitivity: Protect from light.
Fixation: Samples can be stored in IC Fixation Buffer (Product # 00-8222) or 1-step Fix/Lyse Solution (Product # 00-5333) for up to 3 days at 4°C with minimal impact.

Target Information

The CD3 subunit complex transduces antigen-recognition signals into T cells and regulates TCR surface expression. It comprises CD3 gamma, delta, epsilon, and zeta subunits, encoded on chromosome 11. These subunits associate with cytoplasmic signaling molecules via tyrosine-based motifs. CD3 is present on 68–82% of peripheral blood lymphocytes, 65–85% of thymocytes, and Purkinje cells, but not on B or NK cells. Abnormal CD3 expression may be observed in autoimmune diseases. Mutations in CD3 genes can cause immunodeficiency.
