Target Information
Janus kinase 2 (JAK2) is a protein tyrosine kinase involved in cytokine receptor signaling pathways and is mutated in polycythemia vera. JAK2 contains the catalytic domain (JH1) and participates in phosphorylation of STAT proteins, which regulate transcription of interferon-stimulated genes. It is essential for IFN gamma-receptor complex initiation, with JAK1 acting as an amplifier. Mutations in JAK2 are associated with Budd-Chiari syndrome, polycythemia vera, thrombocythemia, myelofibrosis, and acute myelogenous leukemia.
