Product Specific Information
The J43 monoclonal antibody reacts with mouse PD-1 (programmed death-1), a 55 kDa member of the immunoglobulin superfamily. PD-1 contains an immunoreceptor tyrosine-based inhibitory motif (ITIM) and plays a key role in peripheral tolerance and autoimmune disease in mice. PD-1 is mainly expressed on activated T and B lymphocytes. Its ligands, PD-L1 (B7-H1) and PD-L2 (B7-DC), show overlapping functions and are expressed on normal tissues and antigen-presenting cells.
J43 inhibits the binding of mouse PD-L1-Ig and PD-L2-Ig to PD-1/BHK transfected cells. In vivo, J43 enhances contact hypersensitivity and exacerbates acute GVHD, EAE, and NOD diabetes, similar to PD-1 deficiency or PD-L1/PD-L2 blockade.
Applications Reported: Flow cytometric analysis.
Applications Tested: Flow cytometric analysis of stimulated mouse splenocytes at ≤1 µg/test. Optimal titration recommended.
Light Sensitivity: Protect from light due to photo-induced oxidation of tandem dye.
Fixation: Samples can be stored up to 3 days at 4°C in the dark using IC Fixation Buffer (Product #00-8222-49) or 1-step Fix/Lyse Solution (Product #00-5333-54).
Excitation / Emission: 488–561 nm / 775 nm; compatible with Blue, Green, Yellow-Green lasers.
Filtration: 0.2 µm post-manufacturing filtered.
Target Information
PD-1 (programmed death-1, PDCD-1) is expressed on activated T cells and delivers inhibitory signals upon binding to PD-L1 or PD-L2. This interaction limits T-cell activation in the absence of danger signals. PD-1 deficiency leads to hyperactive immune responses and autoimmune diseases. Although predicted at 55 kDa, PD-1 often migrates at a higher molecular weight in SDS-PAGE.
