Target Information
Three isoforms of serine/threonine kinases, designated alphaPAK p68, betaPAK p65 and gammaPAK p62, exhibit high sequence homology with the S. cerevisiae kinase Ste20 involved in pheromone signaling.
These isoforms complex with Rac1 and Cdc42 in their active GTP-bound state, inhibiting GTPase activity and leading to autophosphorylation.
Phosphorylation of Ser141 and Thr402 correlates with gammaPAK activation. Once phosphorylated, the PAK isoforms disassociate to interact with downstream substrates.
