Target Information

Retinoic acid-inducible gene I (RIG-I) is a pattern recognition receptor (PRR) involved in the recognition of viral double-stranded RNA (dsRNA). Along with MDA5, RIG-I detects viral dsRNA and activates the innate immune response. Both are RNA helicases with overlapping and distinct roles.
RIG-I is activated by short dsRNAs lacking a 5′-triphosphate end, while MDA5 responds to long dsRNAs. Once activated, both signal through IPS-1 to activate transcription factors NF-κB and IRF-3, leading to apoptosis, cytokine signaling, and inflammation.
RIG-I is essential for signaling by influenza A/B, human respiratory syncytial virus, paramyxoviruses, Japanese encephalitis virus, and West Nile virus. MicroRNA-146a is involved in feedback inhibition of RIG-I-dependent antiviral response by negatively regulating TRAF6, IRAK1, and IRAK2. Recent findings suggest RIG-I also detects cytosolic DNA through RNA polymerase III activity.
