Cluster of differentiation 8 (CD8) is a type I transmembrane glycoprotein of the immunoglobulin family that plays a critical role in T cell signal transduction, differentiation, and activation.
CD8 is mainly expressed on T cells as a disulfide-linked heterodimer of CD8α and CD8β, functioning as a co-receptor for MHC class I molecules, while CD4 serves as a co-receptor for MHC class II.
CD8α is essential for MHC-I binding and is also expressed on subsets of T cells, NK cells, monocytes, and dendritic cells as homodimers.
Engagement of MHC-I/peptide complexes on antigen-presenting cells triggers Lck kinase activation, leading to lymphokine production, motility, and cytotoxic T lymphocyte (CTL) activation.
Activated CTLs are central to pathogen and tumor cell clearance. Differentiation of naive CD8+ T cells into CTLs is enhanced by IL-2, IL-12, and TGF-β1.
