Target Information

The α/β hydrolase superfamily comprises diverse members involved in key biochemical processes and various diseases. Despite differing sequences and substrates, they share a canonical α/β hydrolase fold and a catalytic triad composed of histidine, acid, and nucleophile residues. Members of this superfamily are often drug targets for diseases such as diabetes, Alzheimer’s disease, obesity, and blood clotting disorders.

  • ABHD1: Metabolizes smoking xenobiotics
  • ABHD2: Participates in atherosclerosis development
  • ABHD4: Involved in alternative synthesis of N-acyl ethanolamines (NAE); acts as a lysophospholipase selective for N-acyl phosphatidylethanolamine (NAPE)
  • ABHD5: Mutations contribute to Chanarin-Dorfman syndrome
  • ABHD6: May play a role in nervous system metabolism and signaling
  • ABHD14A: Possibly involved in granule neuron development
