Product Specific Information
A recommended positive control tissue for this antibody is breast tissue, though other tissues may also serve as positive controls.
This antibody is intended for laboratory professional use in detecting the corresponding protein in formalin-fixed, paraffin-embedded tissues using manual qualitative IHC techniques. It should be used after primary histopathological diagnosis.
Multiple isoelectric variants of calponin have been identified, but only two molecular weight isoforms exist: 34 kDa and 29 kDa. The 29 kDa (I‑calponin) form is mainly expressed in urogenital smooth muscle, while the 34 kDa form is found in vascular and visceral smooth muscle. This antibody reacts with the 34 kDa form in human aortic medial smooth muscle extracts and does not react with fibroblast extracts.
Calponin binds calmodulin, F‑actin, and tropomyosin, playing a role in smooth muscle contraction regulation. It serves as a marker of differentiated (contractile) smooth muscle phenotype.
IHC Use and Controls
- Use on formalin-fixed, paraffin-embedded sections.
- Perform heat-induced or enzymatic antigen retrieval before staining.
- IHC visualization involves sequential application of primary antibody, link antibody, enzyme complex, and chromogenic substrate, followed by washing steps and microscopic evaluation.
- Positive tissue control must be included in each staining run, ideally with both positive and negative elements.
- External positive control tissues should be processed identically to patient samples and selected for weak positive staining to ensure sensitivity monitoring.
- Negative controls (internal or external) should show absence of staining to confirm specificity.
Target Information
Calponin is a thin filament-associated protein involved in smooth muscle contraction regulation. It binds to actin, calmodulin, troponin C, and tropomyosin. Expressed in smooth muscle and myoepithelial tissues, calponin has two isoforms (34 kDa and 29 kDa).
It is also found in myoepithelial cells of benign and malignant breast lesions and can help distinguish collagenous spherulosis (positive) from adenoid cystic carcinoma in breast tissue. Calponin positivity is also observed in adenoid cystic carcinoma of salivary glands.
