Target Information

Members of the RAS subfamily of GTPases function in signal transduction as GTP/GDP-regulated switches cycling between inactive GDP- and active GTP-bound states.
Guanine nucleotide exchange factors (GEFs), such as Repac (RAPGEF5), act as RAS activators by promoting GTP acquisition to maintain the active state.

RAPGEF5 is a 580 amino acid protein expressed mainly in brain and testis, with weaker expression in heart, placenta, lung, pancreas, and small intestine.
It exists as two alternatively spliced isoforms and is conserved in cow, mouse, and rat.
The gene encoding RAPGEF5 maps to human chromosome 7p15.3, a region associated with positive response to erlotinib therapy in non-small-cell lung cancer.
