Product Specific Information

Description:
The UCHT1 monoclonal antibody reacts with human CD3e, a 20 kDa subunit of the TCR complex. The epsilon chain, along with gamma and delta subunits, is essential for TCR assembly, trafficking, and surface expression. CD3 is expressed by thymocytes and all mature T cells. Crosslinking via immobilized UCHT1 triggers intracellular activation and proliferation pathways.

• Flow cytometric analysis.

Applications Tested:

• Pre-titrated and validated for flow cytometric analysis of normal human peripheral blood cells.
• Recommended usage: 5 µL (0.06 µg)/test in 100 µL final volume (10⁵–10⁸ cells/test).

Light Sensitivity:

• Tandem dye is sensitive to photo-induced oxidation. Protect vial and samples from light.

Fixation:

• Samples can be stored in IC Fixation Buffer (Product #00-822-49) or 1-step Fix/Lyse Solution (Product #00-5333-54) for up to 3 days at 4°C in the dark with minimal impact on brightness and FRET efficiency.
• Clone-specific performance should be empirically determined.

Excitation/Emission:

Target Information

The CD3 subunit complex is essential for transducing antigen-recognition signals into T cells and regulating TCR expression. CD3 includes gamma, delta, epsilon, and zeta subunits, encoded on chromosome 11. These subunits contain cytoplasmic tails with tyrosine-based motifs critical for signal transduction. CD3 is expressed on 68–82% of peripheral blood lymphocytes and thymocytes but not on B or NK cells. Defects in CD3 genes can lead to immunodeficiency.
