Losatuxizumab is a biosimilar that targets EGFR.
EGFR (Epidermal Growth Factor Receptor, HER1, ErbB1) is encoded by the EGFR gene located on chromosome 7 in humans. EGFR belongs to the HER/ERbB family of receptor tyrosine kinases, including ERbB2, ERbB3, and ERbB4.
EGFR is a transmembrane receptor; ligand binding (e.g., EGF, TGF-α) induces dimerization and autophosphorylation of tyrosine residues in the cytoplasmic domain. This activation triggers multiple downstream signaling cascades such as Ras–ERK, PI3K–Akt, Jak–STAT, and PKC pathways, promoting cell growth and proliferation.
Phosphorylation at Y1086 allows GRB2 binding, leading to MAPK pathway activation. Upon activation, EGFR undergoes endocytosis and is either degraded or recycled.
Mutations in the EGFR gene are associated with lung cancer. Overexpression or activation of EGFR correlates with poor prognosis and is observed in various tumors including those of the head and neck, brain, bladder, stomach, breast, lung, endometrium, cervix, vulva, ovary, and esophagus.
