The PolarScreen™ Glucocorticoid Receptor Competitor Assay Kit, Red provides a sensitive and efficient method for high-throughput, fluorescence polarization-based screening of potential glucocorticoid receptor (GR) ligands.
This kit utilizes insect cell-expressed human GR and a novel, high-affinity fluorescent glucocorticoid ligand (Fluormone™ GS Red) in a homogeneous, mix-and-read assay format.

This kit produces a red color read-out; a green version is also available. Both kits determine IC50 values of competitive compounds. Color choice depends on the spectral properties of the compounds and available instrumentation.

Assay 작동 원리

GR은 형광 리간드 Fluormone GS Red와 결합하며, 경쟁 화합물이 존재할 경우 GS Red/GR 복합체 형성이 억제되어 편광 값이 감소합니다.
이 편광 값의 변화는 시험 화합물의 GR 결합 친화도를 결정하는 데 사용됩니다.
실험은 상온에서 수행되며, 안정화 펩타이드 사용 시 일관된 결과를 제공합니다.
또한 4°C에서도 안정화 펩타이드 없이 수행 가능하며 GR 결합 특성에 큰 변화가 없습니다.
