Target Information
Cluster of differentiation 8 (CD8) is a type I transmembrane glycoprotein of the immunoglobulin receptor family. It plays an essential role in signal transduction, T cell differentiation, and activation.
CD8 is mainly expressed on T cells as a disulfide-linked heterodimer of CD8α and CD8β, functioning as a co-receptor with the T cell receptor (TCR) for MHC class I molecules.
It is also found on subsets of T cells, NK cells, monocytes, and dendritic cells as CD8α homodimers.
Binding of MHC-I/peptide complexes to CD8 triggers recruitment of Lck kinase, promoting lymphokine production, motility, and activation of cytotoxic T lymphocytes (CTLs).
Activated CTLs are crucial for pathogen and tumor cell clearance. Differentiation of naive CD8⁺ T cells into CTLs is enhanced by IL-2, IL-12, and TGF-β1.
