Target Information

NUDT16 is an RNA-binding and decapping enzyme that catalyzes the cleavage of the cap structure of snoRNAs and mRNAs in a metal-dependent manner. It is part of the U8 snoRNP complex required for the accumulation of mature 5.8S and 28S rRNA.
The enzyme exhibits diphosphatase activity, removing m7G and/or m227G caps from U8 snoRNA and leaving a 5′ monophosphate on the RNA. It also catalyzes cleavage of mRNA cap structures but does not hydrolyze cap analogs like m7GpppG.
NUDT16 has broad substrate specificity with manganese or cobalt as cofactors and binds to U8 snoRNA independently of metal ions. It may regulate snoRNA and mRNA degradation and acts as a phosphatase hydrolyzing non-canonical purine nucleotides (IDP, dITP, GDP, etc.) to their monophosphate forms, preventing incorporation of abnormal purines into nucleic acids.
