Product Description
Norgen’s Urine DNA Isolation Kit for Exfoliated Cells or Bacteria enables rapid isolation of:
- Human genomic DNA from exfoliated cells shed into urine from the urinary tract
- Bacterial genomic DNA from urine samples
The kit isolates DNA from 1–50 mL urine. The purified DNA can be used for diagnostic and research applications, including detection and monitoring of bladder, kidney, or urinary-tract cancers. It also isolates bacterial DNA from both human and animal urine to study bacterial levels and types present. The kit effectively isolates DNA from both Gram-negative and Gram-positive bacteria, including E. coli, Proteus spp., Klebsiella spp., Enterobacter spp., Serratia spp., Pseudomonas spp., Clostridial spp., Leptospirosis spp., Chlamydia trachomatis, and Neisseria gonorrhoeae.
Typical yields vary depending on cell or bacterial density, which is influenced by health, diet, and sex. The kit is sensitive enough to isolate DNA from <10,000 CFU/mL bacteria found in healthy individuals. Purified DNA is fully digestible with all tested restriction enzymes and compatible with PCR, qPCR, and Southern blot analyses.

Storage Conditions and Stability
All solutions should be tightly sealed and stored at room temperature. The kit is stable for 2 years from shipment. Proteinase K is provided in a buffered storage solution and remains stable for up to 2 years at room temperature.

- Few cells or bacteria may be present in urine (<10,000 CFU/mL typical for healthy individuals).
- Incomplete lysis: extend Proteinase K digestion or use fresh samples.
- Incomplete elution: ensure centrifugation at 14,000 RPM after initial spin.
Q2. Why is my genomic DNA sheared?
- Handle samples gently; minimize vortexing (≤15 sec).
- Use fresh urine to prevent degradation by DNases and proteases.
Q3. Why does DNA perform poorly in PCR?
- Inhibitors may be present in urine. Perform an additional wash with Resuspension Solution A + Proteinase K before lysis.
