Target Information
CCR4 is a chemokine receptor preferentially expressed on type 2 helper T (Th2-type) cells. Unlike other chemokine receptors, CCR4 and CCR8 expression on Th2 cells increases transiently following TCR and CD28 engagement. Activated Th1 cells also up-regulate CCR4 expression and responsiveness to thymus- and activation-regulated chemokine.
Chemokines are small (approximately 8–14 kD), mostly basic, structurally related molecules that regulate leukocyte trafficking through interactions with 7-transmembrane, G protein-coupled receptors. They play key roles in immune system development, homeostasis, and function, as well as in the central nervous system and endothelial cell regulation related to angiogenesis or angiostasis.
Chemokines are divided into two major subfamilies:
- CXC chemokines: first two conserved cysteines separated by one amino acid
- CC chemokines: first two cysteines adjacent
