Product Specific Information

The MIH24 monoclonal antibody reacts with human TRANCE (TNF-related activation-induced cytokine receptor), also known as RANKL (receptor activator of NF-kappaB ligand) and OPGL (Osteoprotegerin Ligand).
TRANCE is a type II membrane protein (~35 kDa) closely related to TRAIL, FasL, and TNF. Its mRNA is upregulated by TCR stimulation, and TRANCE-deficient mice lack osteoclasts.
TRANCE plays a critical role in osteoclast differentiation and in T cell–dendritic cell and T cell–B cell interactions. The TRANCE-RANK interaction regulates expression of the anti-apoptotic molecule Bcl-xL.

Target Information

This gene encodes a member of the TNF cytokine family, a ligand for osteoprotegerin, and a key factor in osteoclast differentiation and activation.
It functions as a dendritic cell survival factor and regulates T cell-dependent immune responses.
Activation of T cells induces expression of this gene, promoting osteoclastogenesis and bone resorption.
The protein activates antiapoptotic kinase AKT/PKB through a complex involving SRC and TRAF6, suggesting a role in apoptosis regulation.
Gene disruption in mice leads to severe osteopetrosis, lack of osteoclasts, defects in early T/B lymphocyte differentiation, and impaired mammary gland development during pregnancy.
Two alternatively spliced transcript variants have been identified.
