Target Information
TRPM6 is predominantly expressed in the kidney and colon, encoding a protein that contains both an ion channel domain and a protein kinase domain. It is crucial for magnesium homeostasis and plays an essential role in epithelial magnesium transport and active magnesium absorption in the gut and kidney. Isoforms of M6-kinase lack the ion channel region. Mutations in TRPM6 are associated with hypomagnesemia with secondary hypocalcemia. Alternatively spliced transcript variants encoding different isoforms have been noted.
TRPC7 is thought to form a receptor-activated, non-selective calcium-permeant cation channel operated by a phosphatidylinositol second messenger system activated by receptor tyrosine kinases or G-protein coupled receptors. TRPC7 is also activated by diacylglycerol (DAG) and possibly by intracellular calcium store depletion.
