Target Information
DNA methylation, or the addition of methyl groups to cytosine bases in CpG dinucleotides, is essential for proper development and gene expression regulation. The methylation pattern involves enzymatic methylation and demethylation processes.
MBD4 (also known as MED1) is a mammalian protein with demethylase activity associated with the methyl-CpG-binding domain (MBD). It can revert methylated cytosine bases to cytosines by catalyzing the removal of the methyl group as methanol.
MBD family members (MBD1, MBD2, MBD3, MBD4) are expressed in somatic tissues, with MBD4 showing homology to bacterial DNA glycosylases/lyases involved in base excision repair. In some microsatellite unstable tumors, MBD4 is mutated at an exonic polynucleotide tract.
