Target Information

Peroxiredoxin (Prx) is a growing peroxidase family associated with cell proliferation, differentiation, and apoptosis.
Mammalian Prx proteins (approximately 50 isoforms) are classified based on amino acid sequence homology, particularly in the amino-terminal region containing the active site cysteine residue.
They exhibit thiol-specific antioxidant activity and are distributed across all kingdoms of life.

Mammalian Prx consists of six different members grouped into:

• Typical 2-Cys Prx
• Atypical 2-Cys Prx
• 1-Cys Prx

Except for Prx VI (1-Cys subgroup), the other five 2-Cys Prx isoforms possess thioredoxin-dependent peroxidase (TPx) activity utilizing thioredoxin, thioredoxin reductase, and NADPH as a reducing system.

Mammalian Prxs are 20–30 kDa in molecular size and vary in subcellular localization:

• Prx I, II, VI: Cytosol
• Prx III: Mitochondria
• Prx IV: Endoplasmic Reticulum and secretion
• Prx V: Complex distribution including peroxisome, mitochondria, and cytosol
