Target Information
PAI1 (plasminogen activator inhibitor 1)는 serine protease inhibitor 초과가족에 속하며, 조직형 및 urokinase형 plasminogen activator (tPA 및 uPA)의 주요 억제제입니다. 혈소판은 순환 PAI1의 주요 공급원이며, 섬유아세포, 평활근세포, 내피세포, 간세포, 염증세포 등 다양한 조직과 세포에서 합성 및 분비됩니다. PAI1의 발현은 성장인자, 사이토카인, 호르몬, 염증 인자, 대사물질, 혈관 조절 인자, 화학물질 및 환경적 요인 등에 의해 전사 수준에서 조절될 수 있습니다.
PAI1은 비만, 혈전증, 대사증후군 등 다양한 질환 상태에서 증가된 수준으로 존재하며, 혈전 발생 증가와 관련이 있습니다. PAI1 결함은 비정상적인 출혈로 특징지어지며, fibrinolysis를 억제하고 plasminogen activator의 활성을 저해함으로써 신경세포 생존을 촉진할 수 있습니다. 또한 PAI1 결함은 plasminogen activator inhibitor-1 결핍증의 원인이 될 수 있습니다.
PAI1은 다양한 isoform을 인코딩하는 대체 스플라이싱 전사체 변형이 존재합니다.
