Applied Biosystems™ PureQuant™ Treg Assay

The Applied Biosystems PureQuant assays are a family of qPCR-based assays designed to accurately identify and quantify specific immune cell types within mixed cell populations. These assays measure genomic DNA methylation at unique sites that serve as identifiers for each cell type. Each 96-well plate accommodates up to seven test samples, and results are expressed as the percentage of the target cell type out of total input cells using a free analysis tool.

The PureQuant Treg Assay specifically targets a demethylated region in the FOXP3 gene to identify regulatory T-cells (Tregs). This allows clear discrimination between Tregs and activated CD4+ FOXP3+ conventional T-cells.

  Corrects for bisulfite conversion variation. Plasmid-based calibrator includes genomic target regions and GAPDH in equimolar ratios. Enables estimation of assay-specific bisulfite efficiency factor (TpG/GAPDH ratio).

  Genomic DNA from pooled healthy donors used to verify assay performance. Provides expected range of target cell type abundance.
• Standards:
  Six dilutions covering a 4-log dynamic range (30–31,250 copies) for accurate copy number determination using standard curves.
