Keyhole limpet hemocyanin (KLH) is a large, heterogeneous glycosylated protein composed of subunits with molecular weights of approximately 350,000 and 390,000, forming aggregates of 4,500,000–13,000,000. Each domain of a KLH subunit contains two copper atoms that bind a single oxygen molecule (O₂), giving the molecule a distinctive transparent blue color when oxygenated.

KLH is highly immunogenic yet safe for human use, making it a valuable carrier protein in vaccine development and antibody production. It cannot be synthesized artificially and is purified from the marine gastropod Megathura crenulata.

KLH is widely used as a carrier protein for haptens—small molecules such as peptides, small proteins, and drug compounds that are not immunogenic on their own. The large size and multiple epitopes of KLH induce strong immune responses, and its abundance of lysine residues allows efficient hapten coupling. Its phylogenetic distance from mammalian proteins minimizes false positives in mammalian model research.

Note: KLH may aggregate or precipitate, which can affect hapten conjugation but does not reduce immunogenicity.
