Target Information
Interferon gamma (IFN-γ, Type II interferon) is a macrophage activation factor and immune interferon produced primarily by T-lymphocytes and natural killer cells in response to antigens, mitogens, Staphylococcus enterotoxin B, phytohemaglutanin, and other cytokines.
IFN-γ is a dimeric glycoprotein composed of two 146 amino acid subunits, functioning as a homodimer of approximately 45 kDa. On SDS-PAGE, it appears as 25, 20, and minor 15.5 kDa bands due to differential glycosylation.
The biological activity of IFN-γ is highly species-specific; human IFN-γ does not cross-react with mouse. Its functions include:
- Antiviral and tumor antiproliferative activity
- Induction of class I and II MHC expression
- Macrophage activation
- Enhanced immunoglobulin secretion by B lymphocytes
IFN-γ acts synergistically with other cytokines and signals through IFN-γ receptor I and II, activating the JAK-STAT pathway. It shares about 40% sequence homology with mouse IFN-γ and is upregulated by IL2, FGF basic, and EGF, while downregulated by vitamin D3 or DMN. Mutations in the IFN-γ gene are associated with aplastic anemia.
