Background
- Function: Able to inhibit all four classes of proteinases through a unique “trapping” mechanism. Contains a peptide stretch called the “bait region” with specific cleavage sites for different proteinases. When cleaved, the protein undergoes a conformational change that traps the proteinase. The trapped enzyme retains activity against low molecular weight substrates but loses activity against high molecular weight substrates.
- Similarity: Belongs to the protease inhibitor I39 (alpha-2-macroglobulin) family.
- Subunit: Homotetramer composed of two pairs of disulfide-linked chains.
- Tissue Specificity: Found in plasma; prominent constituent of late-pregnancy sera.
