Target Information

FOXK1 is a transcriptional regulator involved in glucose metabolism, aerobic glycolysis, muscle cell differentiation, and autophagy. It recognizes and binds the forkhead DNA sequence motif (5'-GTAAACA-3'), acting as both activator and repressor depending on context.
Together with FOXK2, FOXK1 regulates metabolic reprogramming toward aerobic glycolysis by promoting glycolytic enzyme expression (HK2, PFK, PKLR, LDH) and suppressing pyruvate oxidation via PDK1 and PDK4.
It participates in gluconeogenesis during fasting and in mTORC1-mediated metabolic reprogramming, influencing genes like HIF1A. FOXK1 also acts as a negative regulator of autophagy in skeletal muscle, repressing autophagy gene expression during starvation.
Additionally, FOXK1 regulates myogenic progenitor cell populations, represses FOXO4-mediated differentiation, and contributes to muscle repair processes. It enhances Wnt/β-catenin signaling by nuclear translocation of DVL and promotes antiviral gene expression by binding ISRE elements.
