Target Information

Ubiquitin-like protein ISG15 plays a key role in the innate immune response to viral infection, either via its conjugation to a target protein (ISGylation) or via its action as a free or unconjugated protein. It is secreted from monocytes in response to type I interferons and promotes NK-cell proliferation and non-MHC-restricted cytotoxicity.
ISG15 synthesis is stimulated by IFN-α, IFN-β, or IFN-ω, but not IFN-γ. Expression is also induced by overexpression of interferon regulatory factors and by influenza B virus.
It is secreted by monocyte, T-lymphocyte, B-lymphocyte, fibroblast, and epithelial cell lines. ISG15 functions as part of the cytokine cascade, amplifying immunomodulatory effects of type I interferons, and also acts intracellularly as a ubiquitin homolog.
Mutations in the ISG15 gene can result in Immunodeficiency 38 with basal ganglia calcification (IMD38).
