Target Information
Programmed death-ligand 2 (PD-L2), also known as B7-DC, is a member of the B7 ligand family within the immunoglobulin superfamily. Along with PD-L1, it acts as a ligand for PD-1. PD-L2 is primarily expressed in dendritic cells but can be induced in various immune and non-immune cells depending on the microenvironment. Its expression is upregulated by IL-4 and to a lesser extent by TNF-α and IFN-γ.
PD-L2 binds PD-1 with higher affinity (2–6×) than PD-L1, functioning as an inhibitory immune checkpoint molecule that regulates T cell activation and cytokine secretion. Interestingly, PD-L2 can also stimulate T cell proliferation independently of PD-1, suggesting additional receptor interactions. One such receptor is repulsive guidance molecule b (RGMb), involved in respiratory tolerance regulation via BMP and neogenin pathways.
