Target Information
CD22 (BL-CAM) is a type 1 integral membrane glycoprotein with a molecular weight of 130–140 kDa. It is expressed in both the cytoplasm and cell membrane of B-lymphocytes and appears early in B-cell differentiation, similar to CD19.
CD22 expression is restricted to mature B cells and is absent in plasma cells, making it useful for phenotyping mature leukemias. It is strongly expressed in hairy cell leukemia, diffuse large B-cell lymphoma, and nodular lymphocyte predominance Hodgkin’s lymphoma, but negative in classical Hodgkin’s lymphoma.
CD22 preferentially binds to α2,6-linked sialic acid, and its recognition site can be masked by cis interactions. Upon ligand-induced tyrosine phosphorylation, CD22 participates in the regulation of B-cell receptor signaling, acting as both a positive and inhibitory regulator through interactions with Src family kinases and phosphatases.
