Background

Defects in MEF2A may cause autosomal dominant coronary artery disease 1 with myocardial infarction (ADCAD1) [MIM:608320].
MEF2A acts as a transcriptional activator binding specifically to the MEF2 element found in various muscle-specific genes and regulates growth factor- and stress-induced genes. It plays roles in skeletal and cardiac muscle development, neuronal differentiation, and survival.

Post-translational Modifications (PTM):

  • Acetylation on Lys-403 activates transcriptional activity; hyperacetylation leads to cardiac myocyte growth and heart failure.
  • Phosphorylation on Ser-408 promotes sumoylation at Lys-403, preventing acetylation.
  • Dephosphorylation by PPP3CA switches sumoylation to acetylation, inhibiting dendrite differentiation.
  • Phosphorylation on Thr-312/319 activates transcription via p38 MAPK signaling.
  • Caspase-mediated cleavage under neurotoxicity leads to neuronal apoptosis.
  • Sumoylation on Lys-403 represses transcriptional activity, facilitated by PIAS1 and SUMO1/2/3.
