  • Catalytic activity: ATP + a [protein]-L-tyrosine → ADP + a [protein]-L-tyrosine phosphate
  • Domains:
    • C-terminal region contains the focal adhesion targeting (FAT) sequence that mediates localization to focal adhesions.
    • The first Pro-rich domain interacts with SH3 domain of CRK-associated substrate (BCAR1) and CASL.
  • Function:
    Non-receptor protein-tyrosine kinase involved in cell motility, proliferation, and apoptosis signaling. Activated by tyrosine phosphorylation in response to integrin clustering, GPCR ligands (e.g., bombesin, lysophosphatidic acid), or LDL receptor occupancy. Plays a role in oncogenic transformation with increased kinase activity.
  • PTM: Phosphorylated on six tyrosine residues upon activation.
  • Similarity: Belongs to the protein kinase superfamily (Tyr protein kinase family, FAK subfamily); contains one FERM domain and one protein kinase domain.
  • Subunit interactions: Interacts with CAS family members, GIT1, SORBS1, BCAR3, RGNEF, SHB, and TGFB1I1.
  • Tissue specificity: Expressed in all organs tested, most abundantly in brain and lymphoid cell lines.
