Background

Defects in TSC2 are associated with lymphangioleiomyomatosis (LAM) [MIM:606690], a progressive lung disease affecting mainly young women, and tuberous sclerosis complex (TSC) [MIM:191100], an autosomal dominant multi-system disorder involving benign overgrowths (hamartomas) and developmental abnormalities (hamartias) in various organs.
TSC2 functions as a tumor suppressor, potentially involved in vesicular transport and cell growth regulation. Interaction between TSC1 and TSC2 facilitates vesicular docking and stimulates intrinsic GTPase activity of RAP1A and RAB5, contributing to growth regulation.
Mutations in TSC2 can lead to constitutive activation of RAP1A in tumors.

  • PTM: Phosphorylation at Ser-1387, Ser-1418, or Ser-1420 does not affect interaction with TSC1.
  • Similarity: Contains one Rap-GAP domain.
  • Subunit Interaction: Interacts with TSC1 and HERC1; stabilized by TSC1 interaction. May also interact with RABEP1, forming a complex linked to RAB5. Interacts with HSPA1 and HSPA8.
  • Tissue Specificity: Expressed in liver, brain, heart, lymphocytes, fibroblasts, biliary epithelium, pancreas, skeletal muscle, kidney, lung, and placenta.
