Background

질병 관련성:
Defects in RB1 are associated with bladder cancer (MIM:109800), osteogenic sarcoma (MIM:259500), and retinoblastoma (MIM:180200). Retinoblastoma is a congenital malignant tumor arising from the retina, occurring in approximately 1 in 20,000 live births and representing about 2% of childhood malignancies. Around 30% of cases are bilateral, and 20% are inherited as an autosomal dominant trait with incomplete penetrance. Diagnosis is usually made before age 2.

기능:
RB1 acts as a key regulator of cell cycle entry and functions as a tumor suppressor. It represses E2F1 target genes, maintaining cell cycle arrest when underphosphorylated. RB1 contributes to heterochromatin formation by stabilizing histone methylation and recruiting histone methyltransferases (SUV39H1, SUV420H1, SUV420H2). It also controls histone H4 Lys-20 trimethylation and inhibits the intrinsic kinase activity of TAF1. Viral proteins such as SV40 large T antigen, HPV E7, and adenovirus E1A disrupt RB1–E2F1 complexes, affecting its regulatory role.
